Trophic actions of oral vanadium and improved glycemia on the pancreatic beta-cell ultrastructure of streptozotocin-induced diabetic rats.

摘要: Context Oral vanadyl sulfate (vanadium) has potent hypoglycemic effects in diabetes animals, but data about its actions on pancreatic beta-cells (BC) ultrastructure is limited. Objective Partial diabetic rats were treated with vanadium and insulin injection their BC are studied. Methods Male made intravenous streptozotocin (STZ, 40 mg/kg). Animals randomly divided to control (CD), vanadium-treated (1 mg/mL VOSO4 + 5H 2 O base solution, VTD) insulin-treated (80 U/kg/day NPH injection, ITD) groups. Treatments started 10 days after STZ terminated months. Intermittent tail blood samples taken for measurements of glucose (BG) plasma (PI). Finally animals sacrificed pancreata prepared assessments ultrastructure, islets histology immunoreactivity (IIR). Results Vanadium decreased BG (P<0.0001), elevated the reduced PI (P<0.001), prevented islet atrophy restored ultrastructure. Low seen during treatment VTD ITD only persisted withdrawal. Hyperglycemia worsened CD repaired shortly atrophied, had scattered IIR spots. pyknotic nuclei, vacuolated cytoplasm few tiny secretory granules. looked normal compact centered well-developed endoplasmic reticulum, many granules mitochondria. structure was slightly better than some immature Conclusion The trophic effectively renovated beta cell atrophy, whereas relief glucotoxicity could repair cells partially atrophy.
