摘要: α-Synuclein aggregation is a hallmark of Parkinson's disease and promising biomarker for early detection assessment progression. The prospect molecular test materializing with the recent developments methods based on amplification synuclein seeds (e.g. RT-QuIC or PMCA). These have shown results identifies synucleinopathies from variety biofluids. However, those experiments are often very long (days) rely precise experimental parameters. Here we adapted single molecule counting aggregates in cerebro-spinal fluid (CSF). This technique performed solution, using approved well-described amyloid reagent thioflavin T dedicated device minimal user input. Single enable to probe events process used ThT-positive aggregates. measurements fast (min) show good sensitivity, down attograms Synuclein when coupled short, isothermal step. Importantly, use also allows refined characterization samples fingerprinting protein present CSF patients. size ThT reactivity each individual aggregate shows unique signature PD patients compared controls may provide new insights future.
