摘要: Abstract Background The aim of this study was to observe the effects neoadjuvant therapy with irinotecan and5-fluorouracil (5-FU)/folinic acid (FA) on resection rate and survival colorectal cancer patients initially unresectable hepatic metastases. Patients methods Forty received chemotherapy comprising 180 mg/m2 administered intravenously (i.v.) day 1, FA 200 i.v. days 1 2, 5-FU 400 bolus 1200 as a continuous 48-h infusion 1. treatment repeated every 2 weeks response assessed 12 (six cycles). Results objective 47.5% (n = 19), two complete responses disease stabilization in 11 (27.5.%) patients. Responses were unconfirmed for undergoing surgery within weeks. Treatment well tolerated adverse events typical agents used. Twenty-seven (67.5%) reported hematological toxicity (35.0% grade 3/4) 14 (35.0%) gastrointestinal (12.5% 3/4). Thirteen (32.5%) underwent potentially curative liver following chemotherapy. Chemotherapy particularly effective large metastases entry study. median time progression is 14.3 months and, at follow-up 19 months, all are alive. Conclusions Neoadjuvant combined 5-FU/FA enabled significant proportion undergo surgical resection. have yet be determined.
