摘要: The immediate effect of tumor necrosis factor (TNF) added to cultures L-cells is cytostasis, manifested as cell arrest in G2. This prevails during the initial 4 hr when number G2 cells increases markedly absence any significant death culture. Shortly thereafter, cytolytic becomes apparent; extensive lysis can be detected after 7 exposure TNF. After 24 nearly all are lysed. Results experiments which effects TNF on kinetics progression through various phases cycle were studied indicate that presence do progress G2, although with considerable delay, and reach mitosis. Most die (undergo lysis) specifically at late stages mitosis (telophase) or soon cytokinesis. Sensitivity lytic increased by Colcemid vinblastine. exerts little rates G1 S cycle, few phase. Nonlysed from treated not show changes nuclear chromatin, suggesting neither DNA nor proteins primary targets present data implicate metabolic occur (cytokinesis), perhaps associated synthesis assembly membrane components, being responsible for sensitivity towards mechanism early cytostatic unknown.
