摘要: Despite its short history, diffraction-unlimited fluorescence microscopy techniques have already made a substantial imprint in the biological sciences. In this review, we describe how stimulated emission depletion (STED) imaging originally evolved, it compares to other optical super-resolution techniques, and what advantages provides compared previous golden-standards for microscopy, such as diffraction-limited electron microscopy. We outline prerequisites successful STED experiments, emphasizing equally critical roles of instrumentation, sample preparation, photophysics, major evolving strategies push borders even further life science.
