Evaluation of human MSCs cell cycle, viability and differentiation in micromass culture.

摘要: Mesenchymal stem cells (MSCs) have the potential to differentiate into distinct mesenchymal tissue cells. They are easy expand while maintaining their undifferentiated state, which suggests that these could be an attractive cell source for engineering of cartilage. In vitro high density micromass culture has been widely used chondrogenesis induction. Our objective was investigate human MSCs cycle, viability and differentiation in conditions. Therefore, induce chondrogenesis, micromasses were cultured presence transforming growth factor-beta1 serum free medium 21 days. Cell phenotype analyzed by flow cytometry. From day 0 7, G0/G1 phase increased, whereas S decreased gradually, but cycle phases (S, G2/M) did not significantly change after 7. Less than 10% apoptotic, no necrosis observed, even at 21. We observed a decrease CD90 CD105 expression, from conclusion, our results demonstrate good during whole period culture. Moreover, allowed synchronized phase, suggested some degree differentiation.
