Comparative, osteochondral defect repair: stem cells versus chondrocytes versus bone morphogenetic protein-2, solely or in combination.

摘要: Full-thickness articular cartilage damage does not resolve spontaneously. Studies with growth factors, implantation of autologous chondrocytes and mesenchymal stem cells have led to variable, some extent inconsistent, results. This work compares osteochondral knee-defect repair in rabbits upon a previously described alginate/(poly(lactic-co-glycolic) acid (PLGA) scaffold distinct conditions. Systems were either vitro pre-cultured small number allogeneic under fibroblast factor (FGF)-2 stimulation or the same amount allogeneic, marrow derived, (without any pre-differentiation), loaded microsphere-encapsulated bone morphogenetic protein (BMP)-2 within alginate layer, holding combinations one other cell type BMP-2. The experimental limit was 12 weeks, because foregoing study this release system had shown maintained tissue response for at least 24 weeks post-operation. After only 6 histological analyses revealed newly formed cartilage-like tissue, which resembled adjacent, normal as well BMP-2 treated defects, but therapy gave higher scores. advantage evened out until weeks. Combinations did result additive synergistic effect. Equally efficient defect achieved chondrocyte, cell, treatment. Expression collagen X I, signs ongoing ossification, histologically undetectable, presence aggrecan indicated tissue. In conclusion, further should demonstrate whether spatiotemporally controlled, on-site alone could become feasible therapeutic approach large defects.
