摘要: To investigate the means by which a cell regulates progression of mitotic cycle, we characterized cdc44, mutation that causes Saccharomyces cerevisiae cells to arrest before mitosis. CDC44 encodes 96-kDa basic protein with significant homology human binds DNA (PO-GA) and three subunits replication factor C (also called activator 1). The hypothesis Cdc44p is involved in metabolism supported observations (i) levels recombination suggest elevated rates damage cdc44 mutants (ii) cycle observed alleviated checkpoint mutations rad9, mec1, mec2. predicted amino acid sequence contains GTPase consensus sites, these regions cause conditional arrest. Taken together, essential gene may encode large subunit yeast C.
