摘要: DNA polymerases I and III are essential for viability of S. cerevisiae. We have cloned analyzed POL2, the gene encoding catalytic subunit third nuclear polymerase, polymerase II. POL2 expressed a transcript approximately 7.5 kb contained reading frame that encoded protein calculated Mr 255,649. The N-terminal half predicted displayed relatively weak similarity sequence to eukaryotic polymerases. Disruption coding at midpoint led viable, slowly growing cells, which yielded truncated polypeptide with II activity, free from subunits B or C. Deletion resulted in inviability dumbbell terminal morphology typically follows arrest replication. conclude three yeast argue all replicases, possibility challenges existing models
