摘要: Summary: Ghrelin is a novel growth hormone (GH)-releasing peptide, isolated from the stomach, which identified as an endogenous ligand for GH secretagogues receptor. Although both ghrelin and its specific receptor are expressed in blood vessels, cardiovascular effects of remain unknown. To clarify whether has vasodilatory effect humans, response forearm flow (FBF) to intra-arterial infusion was examined eight healthy volunteers using plethysmograph. In addition, hormonal responses were studied. increased FBF dose-dependent manner. Pretreatment with NG-monomethyl-l-arginine (l-NMMA), nitric oxide synthase inhibitor, did not inhibit ghrelin, although l-NMMA significantly inhibited release. Serum insulin-like factor-I (IGF-I) level altered by administration. Plasma cyclic guanosine 3´,5´-monophosphate level, second messenger oxide, also altered. These results suggest that possibly through GH/IGF-I/nitric oxide–independent mechanisms.
