摘要: The aim of this study is to investigate the effects molecular weight, pyridinium/trimethyl ammonium (Py/Tr) ratio, nitrogen atoms (N) in methylated N-(3-pyridylmethyl) chitosan chloride (M3-PyMeChC)/the phosphorus (P) DNA (N/P) and physicochemical properties nanopolyplexes on transfection efficiency. water-soluble derivative, M3-PyMeChC, was used as a non-viral vector deliver pEGFP-C2 into human hepatoma (Huh7) cell lines. results revealed that higher weight M3-PyMeChC able form complexes completely with at lower N/P ratios than weights, which led Moreover, Py/Tr showed superior efficiency all studied. highest for occurred 82kDa ratio 5. indicated hydrophobic effect pyridinium moiety could enhance gene efficiency, can be attributed dissociation from nanopolyplexes. High tended decrease cytotoxicity due delocalization positive charge pyridine ring while high increased cytotoxicity. Our spread by delocalizing it heterocyclic ring, suggesting promising approach mediate levels transfection.
