摘要: Allogeneic bone marrow transplantation (BMT) has become a therapy of choice for the treatment certain malignancies and hematopoietic disorders. However, immunodeficiencies following BMT continue to cause significant morbidity mortality. We have compared T cell receptor (TCR) repertoire patients healthy control individuals by staining peripheral blood mononuclear cells with fluorochrome-labeled TCR-specific antibodies. Several exhibited biased pattern TCR expression atypical controls, yet no particular bias characterized all patients. For example, we found that 2%–8% from expressed Vβ19 TCR. One patient on more than 60% cells, while additional less 1% cells. The most extreme skewing types suffered graft-versus-host disease. causes skewed Vβ patterns in are not fully understood, some researchers suggested an oligoclonal expansion CD8+ populations may be largely responsible. To test this hypothesis, examined CD4+ populations. both populations, sometimes within single individual. Thus, therapies directed toward alone correct abnormalities BMT.
