Rate-controlled nano-layered assembly mechanism of melamine-induced melamine–uric acid stones and its inhibition and elimination methods

摘要: The outbreak of infant milk powder poisoning in China attracted worldwide attention. investigations found that the real culprit this incident was melamine (Mel), which induced formation kidney stones. However, little is known about mechanism stones, not to mention preventive and therapeutic drugs. Here, through thermodynamic kinetic tests, simulation vitro animal experiments, multidisciplinary analysis verification, we stone a rate-controlled nano-scale supramolecular layered assembly process: Mel, uric acid (UA) small amount H2O combine hydrogen bonds form planar network structure; structures self-assemble layer-by-layer ellipsoidal nanocrystals π–π stacking interactions, van der Waals force, hydrophilic/hydrophobic interaction, etc.; gradually grow aggregate concentrically Mel–UA stones under control matrix. Based on above process, screen out an effective inhibitor eliminator for potassium citrate (K3Cit), works both bonding alkalinity. This work solves most challenging conundrum left by Mel blazes trail prevention treatment disease.
