Response to 5-fluorouracil chemotherapy is modified by dietary folic acid deficiency in ApcMin/+ mice

摘要: Abstract 5-Fluorouracil (5-FU) has been the foundation of advanced colorectal cancer treatment for over 40 years. The ApcMin/+ mouse, which is genetically predisposed to intestinal neoplasia, was used examine effects 5-FU in this system and impact dietary folic acid on those effects. resulted a 60–80% reduction tumor number. Clinically relevant toxicities, including myelosuppression mucositis, are part response. Tumor numbers rebounded completely following termination therapy, indicating that drug inhibits growth but does not eradicate them. In mice were fed with defined diet containing no (0 ppm), only induced regression pre-existing tumors, also inhibited recovery withdrawal. Our data indicate deficiency, promoting inhibiting recovery, may enhance therapeutic 5-FU.
