Bromodeoxyuridine DNA labelling reveals host and parasite proliferation in a fish-myxozoan model

摘要: Enteromyxum leei is a myxozoan parasite responsible for enteritis in gilthead sea bream (Sparus aurata). The proliferates the paracellular space of intestinal epithelium and induces an inflammatory reaction. To assess cell turnover proliferation, fish were infected with by anal intubation; after 17 64 days, proliferative marker bromodeoxyuridine (BrdU) was administered; 24 hr, tissue samples taken immunohistochemical detection. Parasite exposure induced increased epithelial immune proliferation all segments at time points, even before establishment. This triggered early intubation mainly local level, as shown proliferating nuclear antigen (pcna) gene expression only posterior intestine days (not found lymphohaematopoietic organs). Incorporation BrdU secondary tertiary daughter cells indicated that endogeny not schizogonial division, which uses de novo synthesis pathway pyrimidines. Altogether, immunolabelling pcna showed rapid response intestines upon infection how this effectively reaches or establishes site.
