摘要: Kidney is thought to be a regenerative organ in terms of repair from acute tubular injury. It unknown whether cell population contributes disordered kidney. We attempted identify and isolate highly proliferative cells single cell. dissected nephron adult rat Isolated nephrons were separated into segments cultured. Outgrowing replated after limiting dilution so that each well contained One line which was the most potent grow designated as rKS56. rKS56 showed cobblestone appearance expressed immature markers relating kidney development mature markers. grew exponentially could maintained for 300 days without transformation. In different culture conditions, differentiated defined by aquaporin-1, 2 expression, responsiveness parathyroid hormone or vasopressin. Engrafted ischemic reperfusion model, replaced injured tubules part implantation improved renal function. These results suggest possess character such self-renewal, multi-plasticity capability tissue repair. may possibly contribute future therapy regeneration.
