Alprostadil attenuates myocardial ischemia/reperfusion injury by promoting antioxidant activity and eNOS activation in rats

摘要: PURPOSE To investigate the effect of alprostadil on myocardial ischemia/reperfusion (I/R) in rats. METHODS Rats were subjected to ischemia for 30 min followed by 24h reperfusion. Alprostadil (4 or 8 μg/kg) was intravenously administered at time reperfusion and infarct size, levels troponin T, activity creatine kinase-MB (CK-MB) lactate dehydrogenase (LDH) serum measured. Antioxidative parameters, nitric oxide (NO) content phosphorylated endothelial synthase 3 (p-eNOS) expression left ventricles also Histopathological examinations performed. RESULTS treatment significantly reduced T levels, CK-MB LDH (P<0.05). Furthermore, with decreased malondialdehyde (MDA) (P<0.05) markedly myonecrosis, edema infiltration inflammatory cells. Superoxide dismutase catalase activities (P<0.05), NO level (P<0.01) p-eNOS increased rats treated compared control CONCLUSION These results indicate that protects against I/R injury these protective effects are achieved, least part, via promotion antioxidant activation eNOS.
