ROCK-dependent and ROCK-independent control of cochlear outer hair cell electromotility.

摘要: Outer hair cell electromotility is crucial for the proper function of cochlear amplifier, active process that enhances sensitivity and frequency discrimination mammalian ear. Previous work (Kalinec, F., Zhang, M., Urrutia, R., Kalinec, G. (2000) J. Biol. Chem. 275, 28000-28005) has suggested a role Rho GTPases in regulation outer electromotility, although signaling pathways mediated by these enzymes remain to be established. Here we have investigated cellular molecular mechanisms underlying homeostatic electromotile response guinea pig cells. Our findings defined ROCK-mediated cascade continuously modulates selectively targeting cytoskeleton. A distinct ROCK-independent pathway functions as fast resetting mechanism this system. Neither affects prestin, unique motor These results extend our understanding basic both normal human hearing deafness, revealing key cytoskeleton suggesting ROCK target modulating amplifier.
