NFAT5 Contributes to Osmolality-Induced MCP-1 Expression in Mesothelial Cells

摘要: Increased expression of the C-C chemokine monocyte chemoattractant protein-1 (MCP-1) in mesothelial cells response to high glucose concentrations and/or osmolality plays a crucial role development peritoneal fibrosis during continuous ambulatory dialysis (CAPD). Recent studies suggest that kidney osmolality-induced MCP-1 upregulation is mediated by osmosensitive transcription factor, nuclear factor activated T 5 (NFAT5). The present study addressed question whether activation NFAT5 hyperosmolality, as PD fluids, contributes cell line Met5A. Hyperosmolality, induced addition glucose, NaCl, or mannitol growth medium, increased activity and stimulated Met5A cells. siRNA-mediated knockdown attenuated substantially. Hyperosmolality also factor-κB (NF-κB). Accordingly, pharmacological inhibition NF-κB significantly decreased expression. Taken together, these results indicate osmolalities activate turn upregulates MCP-1, likely combination with NF-κB, thus may participate CAPD.
