摘要: Purpose: The prognosis of head and neck squamous cell carcinomas (HNSCC) remains disappointing the development novel anti-cancer agents is urgently awaited. We identified by a functional genetic screen microRNAs that are selectively lethal for cancer cells but not normal cells. further investigated genes targeted these microRNAs. Experimental Design: A retroviral expression library human was introduced in HNSCC lines oropharyngeal keratinocytes to identify tumor-selective Potential downstream gene targets were profiling validated assays. Results: six inhibit proliferation By 3′-untranslated region (UTR) luciferase reporter assays, we showed ataxia telangiectasia mutated ( ATM ) common target at least two likely three Specific inhibition resulted similar tumor-specific effect, whereas phenotype reverted rescue experiments. Conclusions: These might be developed as highlight an interesting therapeutic cancer. Clin Cancer Res; 19(20); 5647–57. ©2013 AACR .
