Regulated expression of a wheat germin gene in tobacco: oxalate oxidase activity and apoplastic localization of the heterologous protein

摘要: Wheat (Triticum aestivum) germin is a homopentameric glycoprotein whose synthesis allied with seed germination. Germin pentamers show an unusual resistance to dissociation and possess oxalate oxidase (OxO) activity. In order increase our knowledge of gene expression, the function(s) during development possible uses in plant genetic engineering, vivo expression system required. To this end, for germin, named gf-2.8, was studied by expressing either promoter-GUS fusions or intact transgenic tobacco (Nicotiana tabacum) plants. Heterologous transcription monitored vitro GUS OxO activity found occur developing seeds seedlings. This stimulated auxins, as would be expected because presence putative auxin-responsive elements promoter gf-2.8 gene. Auxin stimulation also extended young leaves since could detected treated but not untreated leaves. The biochemical characteristics wheat were conserved host: present under form doublet co-migrating G G' isoforms.
