摘要: Microfabricated, asymmetrical, reservoir-containing particles for use in the intravenous delivery of cytotoxic agents such as melittin to tumors is disclosed. The have a selected shape and uniform dimensions preferably 1 μm 10 range. reservoirs open face particle are filled with solution or suspension therapeutic agent excipients. drug/excipient may be dried by standard techniques. excipients delay dissolution/release from 1-48 hours after rehydrated injected. Alternatively, pore plugged an erodable material covered semipermeable membrane. grafted layer specific ligands designed quickly bind surface either tumor cells vascular endothelial cells, which form capillaries. cytolytic agent, released binding, presented directly membranes target cells. locally high concentration achieved circumscribed volume between juxtaposed cell provides efficient entry into membrane leading lysis ans death. Also disclosed microfabrication methods making method treating cancer patients particles.
