[Expression profiling and functional analysis of hsa-miR-125b and its target genes in drug-resistant cell line of human gastric cancer].

摘要: The expression of hsa-miR-125b is significantly downregulated in the fluorouracil-resistant cell line human gastric cancer (BGC823/Fu). In order to investigate role drug-resistance acquisition process cancer, we performed a series analysis on sequence characteristics, species conservation, target genes, function annotation and signal transduction pathway enrichment using combined bioinformatic approach such as miRbase, TargetScan6.2, PicTar, miRanda, Gene Ontology(GO) KEGG. results showed that miR-125b highly conserved multiple species. A total 79 genes related transcription regulation, protein binding, enzyme activity (P<0.001) were predicted by bioinformatics software. These involved many biological processes including cycle, proliferation, apoptosis responses cytokine, drug DNA damage (P<0.001). And these mainly belong MAPK, Wnt p53 pathways (P<0.01). revealed may regulate pathways, drug-resistant occurrence associated with apoptosis, cycle signaling p53. We suggest affect chemosensitivity regulating above might be reliable candidates for exploring tumor chemoresistance.
