摘要: Background: Hypersensitivity to cross-reactive mannan polysaccharide allergens of saprophytic yeasts is likely be involved in the pathogenesis atopic eczema dermatitis syndrome (AEDS). Mannans induce elevated specific immunoglobulin E and lymphoproliferative responses peripheral blood mononuclear cells (PBMCs). To gain more detailed data involvement different subpopulations PBMCs AEDS after stimulation, changes cell-surface marker distribution were analysed. Methods: The Ficoll-isolated eight yeast hypersensitive patients seven non-AEDS controls stimulated vitro by (CAM) or whole extract antigen [In-House Reference (IHR)] Candida albicans tuberculin [purified protein derivative (PPD)] immunofluorescence staining analysed flow cytometry. expression cytokine mRNA was measured kinetic real-time polymerase chain reaction (TaqMan®). Results: After 7-day there significant increases CD3/CD16+CD56 ratio (P = 0.028 with P = 0.006 IHR), CD4/CD8 (P = 0.049 mannan) interleukin-4/interferon-γ (IL-4/IFN-γ) IHR) a decrease CD3/CD19 (P = 0.035 patients' as compared healthy controls' cells. These not seen cultures PPD. Conclusions: observed CAM IHR-induced elevations T cell/natural killer cell, IL-4/IFN-γ ratios suggest that C. albicans-induced TH2-type can also play role AEDS.
