摘要: To study the effect of changes in plasma non-esterified fatty acid concentration on suppression hepatic glucose production by insulin eight Type 2 (non-insulin-dependent) diabetic patients participated three euglycaemic, hyperinsulinaemic (108pmol.m2-1.min-1) clamp studies combined with indirect calorimetry and infusion [3-3H]-glucose [1-14C]palmitate; (1) a control experiment NaCl 154 mmol/l, (2) heparin was infused together insulin, (3) an antilipolytic agent, Acipimox, administered at beginning experiment. Six healthy volunteers Plasma concentrations during were patients: 151 +/- 36 mumol/l, 949 178 65 9 mumol/l; subjects 93 13 mumol/l. Non-esterified transport rate, oxidation non-oxidative metabolism significantly higher than Acipimox (p less 0.001). Suppression impaired compared to (255 42 vs 51 29 mumol/min, p 0.01). Infusion did not affect (231 49 mumol/min), whereas enhanced (21 53 0.001 mmol/l experiment). We conclude that insulin-mediated is affected increased availability. In contrast, decreased availability enhances insulin.
