摘要: Trypanosoma cruzi is the protozoan parasite that causes Chagas disease or American trypanosomiasis. Kinetoplastid parasites could be considered as model organisms for studying factors involved in posttranscriptional regulation because they control gene expression almost exclusively at this level. The PUF (Pumilio/FBF1) protein family regulates mRNA stability and translation eukaryotes, several members have been identified trypanosomatids. We used a ribonomic approach to identify putative target mRNAs associated with TcPUF6, member of T. family. TcPUF6 expressed discrete sites cytoplasm various stages life cycle not machinery. overexpression tandem affinity purification-tagged allowed identification by purification assays microarray hybridization yielding nine mRNAs. Whole analysis transfected showed were down-regulated populations overexpressing TcPUF6. association TcDhh1 helicase vivo cellular co-localization these proteins epimastigote forms suggest promotes degradation its through interaction RNA complexes. Analysis levels TcPUF6-regulated genes during their transcripts up-regulated metacyclic trypomastigotes. In infective no between was observed. Our results half-lives via differential complexes throughout cycle.
