摘要: Abstract The effect of highly purified recombinant human renin (rh-renin), expressed in Chinese hamster ovary cells, on mean blood pressure (MBP) was evaluated pithed spontaneously hypertensive rats (SHR) and Wistar-Kyoto (WKY). Intravenous bolus injection rh-renin produced dose-dependent increases MBP SHR WKY. pressor response to about 3 times as potent that infusion progressive during the first 40 min, reaching plateaus thereafter maintained up 120 min. This antagonized by inhibitors, YM-21095 KRI-1314, which inhibited reaction between tetradecapeptide competitively, with K i values 5.1 × 10 −10 4.3 −9 M, respectively. In rh-renin-infused SHR, hypotensive 37 KRI-1314. These results suggest can stimulate rat renin-angiotensin system, thereby producing hypertension. Moreover, model could be useful evaluate inhibitor.
