摘要: Highly purified recombinant human renin (rh-renin), synthesized by Chinese hamster ovary cells, was labeled with iodine-125 and given intravenously to pentobarbital-anesthetized common marmosets (Callithrix jacchus) study the fate of circulating renin. Specific anti-rh-renin antiserum used identify 125I-rh-renin. Plasma disappearance exogenously administered 125I-rh-renin in (n = 6) showed two exponential components, a half-life 12.1 +/- 1.9 minutes for rapid component 120.3 16.4 slow component. The metabolic clearance rate 1.17 0.26 ml/min/kg. Thirty after injection 125I-rh-renin, 43.1 0.9 3.5 0.5% injected dose had distributed liver kidneys, respectively. With time, accumulated kidneys decreased. accumulation less than 1% other organs such as lungs, heart, spleen, adrenal glands, testes, ovaries. Analysis kidney extracts high performance liquid chromatography at 30 120 indicated that immunoreactive decreased time accompanied an increase nonimmunoreactive degradation products low molecular weight. incubation monkey or plasma 37 degrees C did not degrade Therefore, rh-renin rapidly cleared from circulation kidney.
