Identification of the nerve terminal targets of botulinum neurotoxin serotypes A, D, and E.

摘要: Botulinum neurotoxins are metalloproteins with one zinc atom bound to the binding motif of endopeptidases. Here we show that botulinum neurotoxin serotypes A, D, and E endoproteases specific for components synaptic vesicle docking fusion complex. Serotypes A cleave SNAP-25, a 25-kDa protein terminal, while serotype D is VAMP/synaptobrevin, membrane vesicles. Both rat brain VAMP isoforms cleaved at single Lys-Leu peptide bond. The proteolytic activity these inhibited by EDTA captopril.
