Efficient isolation and proteomic analysis of cell plasma membrane proteins in gastric cancer reveal a novel differentiation and progression related cell surface marker, R-cadherin

摘要: Cell plasma membrane proteins, playing a crucial role in cell malignant transformation and development, were the main targets of tumor detection therapy. In this study, CyDye/biotin double-labeling proteomic approach was adopted to profile proteome gastric cancer line BGC-823 paired immortalized epithelial GES-1. Real-time PCR, Western blotting, immunohistochemical staining used validate differential expression novel identified surface marker R-cadherin cells tissues. Clinicopathological study survival analysis performed estimate its roles progression outcome prediction. PCR blotting showed that level significantly lower than non-cancerous indicated dominantly expressed on normal epithelium, deletion tissues associated with site, differentiation, lymph node metastasis, pTNM (chi-square test, P < 0.05). Those patients positive displayed better overall survivals negative group (log-rank P = 0.000). Cox multivariate revealed lacking independent predictor for poor clinical (RR = 5.680, 95 % CI 2.250–14.341, P < 0.01). We have established fundamental database as differentiation progression-related cancer. Lacking indicates prognosis
