摘要: Mitogen-activated protein kinases such as Erk1 and Erk2 serve a paradigm for growing family of proline-directed that mediate entry, progression exit from the cell cycle in diverse eukaryotic cells. These enzymes function within highly conserved modules sequentially activating transduce signals extracellular stimuli. In vertebrates, at least three distinct have been characterized. Mitogens induce sequential activation Raf1→Mek1→Erk2→Rsk via G-protein Ras. Stress factors stimulate c-Jun through related kinase pathway involving Mekk→Sek→SAPK c-Jun, hsp27 phosphorylation MKK3→Hog→MAPKAPK-2 route. Genetic biochemical studies, example budding yeast, imply existence several can operate parallel or integrated systems.
