Comparison of the early response of human embryonic stem cells and human induced pluripotent stem cells to ionizing radiation

摘要: Despite the well-demonstrated efficacy of stem cell (SC) therapy, this approach has a number key drawbacks. One important concern is response pluripotent SCs to treatment with ionizing radiation (IR), given that used in regenerative medicine will eventually be exposed IR for diagnostic or treatment‑associated purposes. Therefore, aim present study was examine and compare early IR‑induced responses assess their radioresistance radiosensitivity. In study, 3 lines; human embryonic (hESCs), induced (hiPSCs) primary dermal fibroblasts (PHDFs); were at doses ranging from 0 to 15 gray (Gy). Double strand breaks (DSBs), gene expression following DNA repair genes analyzed: P53; RAD51; BRCA2; PRKDC; XRCC4. hiPSCs demonstrated greater radioresistance, as fewer DSBs identified, compared hESCs. Both SC lines exhibited distinct profiles most common are involved homologous recombination, non‑homologous end‑joining enhanced damage exposure. Although hESCs equivalent terms capacity pluripotency differentiation into germ layers, results indicate these 2 types differ exposure IR. Consequently, further research required determine whether equally safe application clinical practice. The contributes understanding (DDR) mechanisms activated may aid future development SC‑based protocols.
