摘要: The effects of glimepiride, the newest sulphonylureic compound, on pancreatic insulin and glucagon secretion were studied using classical, isolated, perfused rat pancreas model. influence four different environmental glucose conditions (during a glycaemic stimulus with increasing from 5 to 8.33 mM at stable 0, 2.22 levels) glimepiride was also assessed. At pharmacological concentration strongly stimulated beta-cell activity, producing characteristic biphasic release sharp first-phase secretory peak, followed by prolonged sustained second phase. Environmental concentrations markedly influenced extent, but not pattern glimepiride-induced secretion, as hormone dropped significantly when level reduced. Glimepiride failed alpha-cell activity any levels.
