摘要: Sympathetic lumbar chains from 7 to 15 day old chick embryos were dissociated with consistently greater neuronal and non-neuronal cell yields when the source tissue was collected in Ca++, Mg++− free solutions rather than standard balanced salt or culture media. The 11-day embryonic material used for further studies on dissociation procedure a quantitative analysis of nerve growth factor-supported, dispersed cultures derived it. Neuronal elements attached collagen-coated surfaces different time course, providing way fractionate sympathetic dissociate into neuron-free population rapidly growing cells an over 90% pure overall about 10000 neurons per ganglion. From unfractionated harvests, 25% seeded dish: these, 60% developed processes within first day, survived several days underwent complete reaggregation number masses interconnected by fibre tracts overlying population. In contrast, purified fraction, while yielding initially same neurons, led containing fewer fibre-bearing none them surviving beyond 48 h. inability survive shown be direct consequence absence cells. Seeding increasing numbers pre-attached progressive increase fibre-bearing, long-term decline their tendency coalesce reaggregates.
