Evaluation of spheroid head and neck squamous cell carcinoma cell models in comparison to monolayer cultures

摘要: Two-dimensional (2D) monolayer cell culture models are the most common method used to investigate tumor cells in vitro. In few last decades, a multicellular spheroid model has gained attention due its adjacency tumors vivo. The aim of present study was immunohistochemical differences between these two systems. FaDu, CAL27 and SCC25 head neck squamous carcinoma (HNSCC) lines were seeded out spheroids. FaDu treated with increasing doses cisplatin irradiation. not theproliferation experiments, since spheroids able process reagent CCK-8 assays. Furthermore, they stained alterations following antigens: Ki-67, vascular endothelial growth factor receptor, epithelial survivin. Differences rates expression patterns detected certain HNSCC lines. proliferation showed significant divergence grown three-dimensional compared 2D model. Overall, promising reproduce aspects characteristics cells, may show different response therapeutic options.
