摘要: The phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX) showed inductive effect on larval settlement of the polychaete Hydroides elegans. It has been suggested that IBMX triggers by elevating cellular adenosine 3 ',5 '-cyclic monophosphate (CAMP) level in this species. To test hypothesis, we first examined cAMP-level changes both competent (CL) and attached larvae (AL) then characterized cAMP-dependent protein kinase H. elegans, which is major mediator cAMP action. Tissue extracts were assayed for enzyme immunoassay; results increased production up to approximately two-folds CL. However, there was no significant difference concentration between CL AL not treated with IBMX. catalytic subunit A gene from elegans (designated HePKAc) cloned, its expression different developmental stages using quantitative real-time polymerase chain reaction. pre-competent trochophore lowest, CL, reached highest undergoing normal IBMX-induced metamorphosis, decreased adult stage.
