Localization of sterically stabilized liposomes in Klebsiella pneumoniae-infected rat lung tissue: influence of liposome characteristics.

摘要: Sterically stabilized liposomes are able to localize at sites of infection and could serve as carriers antimicrobial agents. For a rational optimization liposome localization, the blood clearance kinetics biodistribution differing in poly(ethylene glycol) (PEG) density, particle size, bilayer fluidity or surface charge were studied rat model unilateral pneumonia caused by Klebsiella pneumoniae. It is shown that all preparations preferentially infected lung compared contralateral non-infected lung. A reduction PEG density rise size resulted higher uptake mononuclear phagocyte system, lower circulation time localization. Differences did not affect localization Increasing amount negatively charged phospholipids kinetics, but reduce this preparation site infection. In conclusion, degree remarkably independent physicochemical characteristics liposomes. Substantial selective can be achieved provided certain criteria regarding inclusion met. These properties seem direct consequence presence polymer coating operating repulsive steric barrier opposing interactions with biological components.
