摘要: Flow cytometric estimation of nuclear DNA content was performed in six plant species employing three fluorochromes showing different base preferences: propidium iodide (no preference), 4′,6-diamidino-2-phenylindole (DAPI; AT and mithramycin (GC preference). Nuclei isolated from human leukocytes were used as a primary reference standard. While contents estimated using agreement with published data obtained other techniques, the values preference significantly different. It found that differences caused by overall AT/GC ratios, species-specific binding these to DNA. concluded estimations should be interpreted caution even when ratios sample are equal. The use intercalting dyes (e.g. iodide) is recommended for this purpose. On hand, comparison staining behaviour intercalating may give additional information on chromatin structural arrangement molecule pairs
