摘要: Background: Formalin fixed and paraffin wax embedded tissues of necropsy origin are an important source for molecular analysis especially in rare diseases, neuropathology, or epidemiology studies. Because DNA degradation, only short sequences can be amplified from this type tissue, very often less than 100 bases. This poses problems because studies on polymorphism mutations occurring large genes require the long sequences. Methods: The development a simple treatment to obtain longer fragments archival postmortem tissues. Results: It was possible amplify ranging up 300 bases tissues, with no modification usual extraction procedures. To stretches DNA, pre-PCR restoration required, by filling single strand breaks, followed vigorous denaturation step. Conclusions: allowed obtained
