摘要: The bacterium Escherichia coli is the most widely used expression host for overexpression trials of membrane proteins. Usually, different strains, culture conditions and regimes are screened to identify optimal strategy. However, yields often not satisfactory, especially eukaryotic This has initiated a revolution protein in bacteria. Recent studies have shown that it feasible (i) engineer or select E. strains with strongly improved characteristics, (ii) use bacteria other than proteins, (iii) variants retain functionality but express better wild-type protein, (iv) proteins using coli-based cell-free systems.
