摘要: Most cell culture and in vitro drug sensitivity assays utilize serum-supplemented media (SSM). However, fully defined serum-free (SFM) offer several advantages are being used increasingly for initiation maintenance of cultures. Because serum inhibits the cytotoxicity certain antineoplastic agents, we investigated inter-relationships between medium type, proliferation cytotoxic effect. Twenty-four human lung cancer lines were tested with nine anticancer agents both types. A semi-automated tetrazolium (MTT) colorimetric assay was assaying survival. Cell initiated maintained SFM preferentially proliferated that type or equally well In contrast, established SSM varied considerably their preference. There appeared to be a direct correlation trend proliferative rate all drugs possible exceptions methotrexate carmustine. general, more sensitive when they exposed which proliferated. Therefore, determine influence on cytotoxicity, analyzed data only from replicated efficiently media. After correction rate, had negative effect action some (especially methotrexate, 5-fluorouracil and, lesser extent, mitomycin-C). Our results demonstrate may suitable initiating use selection individualized therapy screening new anti-neoplastic thus increase number can satisfactorily.
