摘要: The pharmacological effects of Oren-gedoku-to (OGT), a Japanese-Chinese traditional herbal medicinal mixture on lipid biosynthesis were investigated in cultured human hepatocyte HepG2 cells. addition OGT (0.5 and 4.2 mg/ml), which had no effect cell proliferation cellular protein content, caused marked decrease the cholesterol particularly cholesteryl ester content following 24 h incubation. incorporation 14C-oleate into fraction was also reduced remarkably during incubation for 6 h. OGT, its components main active chemicals acyl-coenzyme A:cholesterol acyltransferase (ACAT) activity studied vitro to explore mechanism by inhibits formation. data confirmed that dose-dependent manner, (Scutellaria baicalensis, Coptis japonica, Gardenia jasminoides Phellodendron amurense) inhibit ACAT activity. Among baicalein, kind flavonoid, decreased fashion from level 10(-6)M. These results strongly suggest reduces formation hepatocytes inhibiting ACAT, baicalein may, part, be responsible inhibition.
