摘要: Incomplete epigenetic modification is one of important reasons inefficient reprogramming the donor cell nuclei in ooplasm after somatic nuclear transfer (SCNT). It may also underlie observed reduced viability cloned embryos. Sodium butyrate (NaBu) a natural histone deacetylase inhibitor that produced intestine. In current study, we evaluated effects NaBu on preimplantation development, acetylation, and gene expression porcine SCNT Our results showed blastocyst rate (24.88 ± 2.09) embryos treated with 1.0 mM for 12 hr activation was significantly higher (P < 0.05) than untreated (13.15 3.07). addition, displayed global acetylated H3 at lysine 14 profile similar to vitro fertilized (IVF) during development. Lower levels Oct4 Bcl-2, but Hdac1, two-cell stages were observed, compared those IVF counterparts. The four-cell no differences these genes among or without NaBu; however, Dnmt3b different. NaBu-treated Oct4, as blastocysts. These indicated treatment alters their acetylation pattern provide beneficial developmental competence expression.
