Preliminary characterization of Lactococcus garvieae bacteriophage isolated from wastewater as a potential agent for biological control of lactococcosis in aquaculture

摘要: Lactococcosis, a significant emerging disease of fish caused by Lactococcus garvieae, has become one the devastating problems due to its serious economic damage in aquaculture. The aim this study was isolate and characterize lytic phage infecting L. garvieae as potential bioagent for treatment lactococcosis. In regard, strain isolated from diseased rainbow trout, then, following biochemical molecular identifications, specific phage, WWP-1, which able destroy cells through cycle, municipal wastewater sample. Transmission electron microscope revealed that possesses an icosahedral head non-contractile short tail, resembled members family Podoviridae. Moreover, WWP-1 represented optimal antibacterial activity at temperatures ranging 15 30 °C, suggesting it could be very effective trout rearing temperature. Restriction profile analysis NdeI can digest genome while EcoRI, EcoRV, BamHI were incapable cutting DNA. According vivo experiment result, decrease mortality rate infected results suggest naturally occurring bacteriophage considered promising agent control strains rearing.
