摘要: Schizosaccharomyces pombe, also known as fission yeast, is an established model for studying chromosome biological processes. Over the years, research employing yeast has made important contributions to our knowledge about segregation during meiosis, well meiotic recombination and its regulation. Quantification of frequency not a straightforward undertaking, either requiring viable progeny genetic plating assay, or relying on laborious Southern blot analysis intermediates. Neither these methods lends itself high-throughput screens identify novel factors. Here, we establish visual assays Sz. pombe characterizing phenotypes. Genes expressing red, yellow, and/or cyan fluorophores from spore-autonomous promoters have been integrated into genomes, close centromere 1 monitor segregation, arm 3 form interval at which can be determined. The assay allows immediate assessment outcome single meiosis by epi-fluorescence microscopy without tetrad dissection. We demonstrate that automatized utilizing imaging flow cytometry enable screens. These several advantages over traditional analyzing
