Therapeutic effect of suicide gene-transferred mesenchymal stem cells in a rat model of glioma.

摘要: We evaluated a new therapeutic strategy for malignant glioma, which combines intratumoral inoculation of mesenchymal stem cells (MSCs) expressing cytosine deaminase gene with 5-fluorocytosine (5-FC) administration. For in vitro and vivo experiments, MSCs were transfected adenovirus carrying either enhanced green fluorescent protein (AdexCAEGFP) or (AdexCACD), to establish MSC-expressing EGFP (MSC-EGFP) CD (MSC-CD). Co-culture 9L glioma MSC-CD medium containing 5-FC resulted remarkable reduction cell viability. The migratory ability MSC-EGFP toward was demonstrated by double-chamber assay. the study, rats harboring brain tumors inoculated MSC-CD. Immunohistochemistry rat showed distribution MSC-EGFP. Survival analysis bearing gliomas treated intraperitoneal significant prolongation survival compared control animals. In conclusion, molecular therapy combining suicide as targeting vehicle represents potential approach both respect antitumor this system its neuroprotective effect on normal tissue.
