摘要: Differentiation-inducing factors (DIFs) are required for stalk cell formation in Dictyostelium discoideum. In the present study, order to support our hypothesis that DIFs may function via increases [Ca2+]c and [H+]c, we investigated combined effects of 5,5-dimethyl-2,4-oxazolidinedione (DMO, a [H+]c-increasing agent), thapsigargin (Tg) BHQ ([Ca2+]c-increasing agents) on vitro several strains. DMO, combination with Tg or BHQ, induced DIF-deficient mutant HM44. Although rates induction by drugs were low presence cerulenin (an inhibitor endogenous DIF production) HM44 V12M2 (a wild-type strain), succeeded inducing sufficient when small amount DIF-1 was supplied. Furthermore, co-addition also AX-4 axenic strain) HM1030 (dmtA−) but not CT15 (dimA−). The suppressed spore promoted both HM18 sporogenous mutant) 8-bromo-cAMP-stimulated V12M2. results suggest function, at least part, [H+]c D.
