摘要: The isolated working rat heart is a useful experimental model which allows contractile function to be measured in hearts perfused at physiologically relevant workloads. To maintain these high workloads the required generate tremendous amount of energy. In vivo this energy derived primarily from oxidation fatty acids. many situations it desirable perfuse presence concentrations This particularly important when studying metabolism heart, or determining how acids alter outcome myocardial ischemic injury [1, 2]. other major source for carbohydrates (glucose and lactate), with smaller ATP also being glycolysis. Two byproducts both acid carbohydrate are H2O CO2. By labeling glucose, lactate, perfusate 3H 14C experimenter can quantitatively collect either 3H2O 14CO2 produced by heart. using radioisotopes that labeled specific hydrogen carbon molecules on various substrates, knowing activity radiolabeled substrate used, possible determine actual rate flux through individual pathways. paper will describe protocols directly measuring hearts.
