Lidocaine inhibits granulocyte adherence and prevents granulocyte delivery to inflammatory sites.

摘要: Incubation of whole blood or PMNs in saline with 10–2 10–4 molar concentrations lidocaine causes a reversible inhibition granulocyte adherence. A single bolus dose lidocaine, 2.5 mg/kg, given intravenously inhibits adherence rabbits by 55.4% at 5 min, return to normal 15 min after dose. When the is followed an infusion 0.3 mg/kg/min, stays less than 50% control values for duration infusion. Plasma from animals granulocytes, and inhibiting factor removed completely dialysis plasma against modified Hanks’ solution 24 hr. Lidocaine prevents delivery sites inflammation: sterile peritonitis developed exudates 6 hr containing mean 19,280 PMN/cu mm compared 388/cu lidocaine-infused (2% control). By comparison, methylprednisolone-treated exudate PMN counts 40% control. was induced staphylococci, died, whereas only 1 noninfused died. Pathologic examination experimentally infected skin showed that inflammatory reaction characterized edema pronounced infiltration 12 intracutaneous injection live staphylococci. In contrast, biopsies edema, virtually no infiltration. Thus, vitro vivo markedly suppresses inflammation.
