Ontogenetic Changes in the Expression of Estrogen Receptor α and β in Rat Pituitary Gland Detected by Immunohistochemistry

摘要: The physiological effects of estrogen on the pituitary, including cellular proliferation and regulation hormone synthesis, are mediated by nuclear receptor (ER). purpose this study was to determine ontogenetic expression two types ERs (ERalpha ERbeta) in pituitary using specific antibodies, monoclonal antibody (1D5) for ERalpha polyclonal generated against ERbeta. First, we confirmed detection 66- 55-kDa bands ERbeta, respectively, rat extract Western blotting. Then immunostaining with these antibodies performed fetal adult Wistar tissues, combined PRL or LHbeta immunohistochemistry. Intense ERbeta signal detected throughout from day 12 gestation. However, staining only became detectable 17 In contrast period, nuclei stained were widely distributed anterior lobe rat, whereas ERbeta-positive cells restricted lobe. LHbeta, but not PRL, colocalized cells. Our results indicated that major population ER subtypes gland has changed around birth may be involved differentiation cell function synthesize a hormone.
